- 01HD0T0DJZG9AQ7ZDENC8R41BS abstract "In Westernized societies, food allergies are a growing epidemic and are considered a major cost to our healthcare system. IgE-mediated cow’s milk allergy (IgE-CMA) is one of the first allergies to arise in early childhood and may occur to various milk allergens, of which beta-lactoglobulin (BLG) and casein are the most important. Understanding the underlying mechanisms behind IgE-CMA is imperative for the discovery of novel biomarkers and the design of innovative treatment and prevention strategies. For this purpose, we sensitized 2 mice strains (BALB/c and C57Bl/6) to BLG using 2 protocols (cholera toxin (CT) or an oil emulsion; n = 6 per group; 120 in total). After sensitization, mice were challenged orally, their clinical signs monitored, antibody (IgE and IgG1) and cytokine levels (IL-10 and IFN-γ) measured, and fecal samples subjected to metabolomics using UHPLC-Q-Orbitrap-HRMS. Jejunum and ileum were collected for histological evaluation. The results of the murine models were further supported by fecal microbiome-metabolome data from our population of allergic (IgE-CMA; n=24) and healthy (n=23) children, on which polar metabolomics, lipidomics and 16S rRNA metasequencing were performed. In vitro gastrointestinal digestions corroborated the microbial origin of proposed metabolic changes. Following antigen challenge, anaphylaxis was observed in BALB/c mice receiving BLG and CT, next to higher levels of antibodies and lower levels of IFN-γ. Changes in microbial metabolites, i.e., secondary bile acids and tryptophan metabolites were observed early on during sensitization, followed by markers of allergic inflammation, i.e. histamine, sphingosine and sphinganine in the later stages. In allergic children on an elimination diet, chronic low-grade inflammation was reflected by dysregulation in tryptophan, bile acid, nucleotide and energy metabolism. Microbial dysbiosis was confirmed by differences in α and ß diversity (p < 0.05), correlation analyses, in vitro digestions, and multi-omics. Our results suggest that metabolic alterations in BALB/c mice sensitized with BLG in CT most closely resemble IgE-CMA in children. We hypothesized that microbial dysbiosis precedes allergic inflammation, with the former aggravating sensitization, as both the gut microbiome and its metabolites take part in immune development.".
