- aggregation abstract "Germ-free <i>Artemia </i> nauplii were fed daily with Baker’s yeast (wild type strain) and dead <i>Aeromonas hydrophila </i> and named group WTY. The other group was fed daily with yeast mutant strain, mann9, and dead <i>Aeromonas hydrophila </i> (group mnY). For histology, two live nauplii per group were fixed at days 2, 4 and 6 in 100% ethyl alcohol, 40% formaldehyde and acetic acid for 5 min, dehydrated in graded alcohols and embedded in paraffin. Five µm thick sections were stained with haematoxylin and eosin and subsequently examined by light microscopy. For TEM, nauplii were fixed overnight in Karnovsky’s fixative, post-fixed overnight in 1% osmium tetroxyde and embedded in SPURR’s resin. Blocks were sectioned at 60 nm and sections were mounted on formvar coated grids and post-stained with uranyl acetate and lead citrate. They were examined using a Jeol 1200 EXII TEM at 80kV accelerating voltage. On day 2 no significant morphological differences in the mid- and hindgut could be observed between both groups. However, on days 4 and 6, significantly longer mid- and hindguts with larger diameters were seen in nauplii of the MnY group. Mnn9 yeast mutant strain sustains better growth under the described culture conditions. This could be due to high amounts of cell-wall chitin and glucans in combination with low amounts of mannose present in mnn9 yeast strain. Since <i>Artemia </i> exhibits low mannase activity in the digestive tract, diets with less mannose content make digestion easy. Furthermore, high levels of chitin and glucan make the diet more nutritious.".
